Abstract

Sugarcane mosaic disease caused by the Sugarcane Mosaic Virus (SCMV) is one of the main sugarcane viruses. The use of resistant cultivars is the most efficient mean of control. The cDNA-AFLP approach has being used to detect genetic polymorphism between contrast phenotypes under controlled conditions. Such polymorphisms can be used to the identification of candidate genes and also to marker development for genetic mapping. The objective of the present work was to identify potential candidate genes related to mosaic resistance using the cDNA-AFLP technique. Two sugarcane cultivars IACSP95-5000 (resistant) and IAC91-1099 (susceptible) obtained through tissue culture will be inoculated with a strong strain of the virus (Rib-1) at green-house condition with three biological replicates for each sample times (0, 24, 48 and 72 hours after inoculation) and respective controls (non-inoculated plants). After inoculation the leaves of each cultivar will be collected at each sample time to the identification of the differentiated expressed fragments revealed through cDNA-AFLP. These fragments, after cloning and sequencing for homology identification could be applied to the development of a primer set with potential use in genetic mapping of sugarcane mosaic resistance regions.
