Abstract

RNA interference mechanism is an event of gene repression through the degradation of messenger RNA. By this way is possible to suppress the target gene expression, so siRNA is a promising therapy for the treatment of severe diseases, including cancer. However, for its effective therapy is necessary to develop delivery systems capable of releasing the siRNA into target cells, promote their cellular internalization and prevent its degradation. For cancer treatment, where there is high cell proliferation and the siRNA effect is for a few days, it is interesting to develop systems that promote a sustained release. An interesting delivery system with this characteristic is the lipid or amphiphilic-based liquid formulation that in situ gelling by self-assembling mechanism, in this case shown by water absorption of the biological environment in the administration site. These systems were developed in the master's project of Livia N. B. Cardoso and were able to form complexes with siRNA, being biocompatible and biodegradable. Thus, this project aims to standardize the obtainment of liquid crystalline systems with in situ gelling, in vitro evaluate of siRNA release profile and cell transfection and to determine in vivo the release profile of siRNA and the effectiveness of these systems in the treatment of squamous cell carcinoma after intratumoral administration. So, this project is a new proposal for gene therapy of skin cancer.
