Effects of early treatment with low level laser therapy on the muscle skeletal repair

Abstract

The low level laser therapy (LLLT) is widely used in order to promote faster and better repair muscle with reference to different types of injury, however, studies on the prior application of the laser are scarce. The aim of this study is to evaluate the effect of LLLT prior to injury on the repair process and in particular on the expression of myogenic regulatory factors MyoD and myogenin and the matrix metalloproteinase MMP-2. For that, will be used 100 Wistar rats divided into 05 groups: (G1) Control, (G2) Sham; (G3) LLLT immediately prior to muscle injury TAD; (G4) No lesion with LLLT immediately prior to sacrifice; (G5) Only injury without treatment with LLLT. The crioinjury applications consist of two rod cooled in liquid nitrogen directly into the TA muscle. Groups 3-5 will be assessed at 1, 3 and 7 days after injury. The LLLT will be performed using the GaAlAs laser (» 780nm) with the density energy of 5 J/cm², output power of 20 mW and time of 10 seconds. For the analysis of expression of myogenic factors will be extracted total RNA from TA muscle and will get the cDNA for the realization of real-time PCR using primers specific for each factor cited and MMP2. To assess the activity of MMP2 is used technique for detecting zymography gelatinase activity in the different groups. (AU)
