Effects of early treatment with low level laser therapy on matrix metalloprotease 2 activity during skeletal muscle repair

Abstract

Low-level laser therapy (LLLT) is widely used in order to promote faster and better muscle repair following different types of injury, however, studies on the previous application of the laser are scarce. The aim of this study is to evaluate the effect of LLLT prior to injury on the matrix metalloproteinase MMP-2 activity in the tibialis anterior (TA) muscle of rats following cryoinjury. Male Wistar rats will be randomly divided into seven groups: (G1) Control, (G2) Sham; (G3) LLLT realized two hours prior to injury in the right TA muscle (D); (G4) LLLT realized immediately prior to muscle injury TAD; (G5) without injury and LLLT realized two hours prior to sacrifice; (G6) without injury and LLLT realized immediately prior to sacrifice; (G7) Only injury without LLLT. The cryoinjury applications consist of two rods cooled in liquid nitrogen directly into the TA muscle. Groups 3-7 will be assessed at 1, 3, and 7 days after injury. The LLLT will be performed using the GaAlAs laser (» 780nm) with the density energy of 5 J/cm², the output power of 20 mW, and the time of 10 seconds. MMP2 activity will analyze by zymography and its presence will be confirmed by Western Blotting.(AU)
