Abstract

All eukaryotic cells are able to store lipid in dynamic and multifunctional structures called lipid droplets (LDs). Structurally there is a lipid core covered by phospholipids and various proteins, and adipophilin is the main one. The increased lipogenesis, which is an important phenomenon for tumor independence, has been observed in various human carcinomas such as colon, renal, hepatic, among others. The lipogenic profile evidenced by the accumulation of cytoplasmic LDs has been implicated with tumor phenotype and biological behavior. The presence of LDs in salivary gland carcinomas is already known in some subtypes with sebaceous differentiation as sebaceous carcinoma and sebaceous linfadenocarcinoma. However, even in the absence of sebaceous differentiation, LDs have been detected in other salivary tumors such as acinic cell carcinoma. Historically, the identification of intracellular lipid was feasible only through the use of fresh tissue and/ or ultrastructural studies. Nevertheless, currently, adipophilin protein, which covers the LDs, can be detected in formalin fixed paraffin embedded tissue by immunohistochemistry. The aim of this study is to analyze the immunohistochemical expression of adipophilin in different histologic subtypes of salivary gland tumors to investigate the frequency of cytoplasmic LDs and their possible association with morphology, degree of differentiation and proliferation.
