Abstract

The essential oil of Melaleuca alternifolia (TTO) is an herbal antifungal action. This study will evaluate the efficacy of TTO and its main component Terpinen-4-ol against resistant clinical isolates and reference strains of Candida albicans and its effect on biofilm developed in acrylic resin blocks. This study will realized in three stages: 1. Identification of the MIC (Minimum Inhibitory Concentration) and MFC (Minimum Fungicidal Concentration) of TTO and Terpinen-4-ol in the planktonic form of C. albicans at the concentrations to be determined before the experiment. Analysis of different concentrations of oil on biofilm SSD (monoespécie) of C. albicans developed in the microtiter plate. Nystatin will be used as control. The microorganisms will be quantified in CFU / ml and assessed for metabolic activity of cells by the XTT method. 3. Biofilm formation developed in specimens of acrylic resin that will be submitted to the following solutions: TTO and Terpinen-4-ol (only in inhibitory concentration determined in the previous phase), PBS (positive control) and nystatin (Sigma). After this test, analyzes will be performed by XTT test and CLSM (confocal microscopy) to determine viable and non viable cells in the biofilm.
