Abstract

The variability and the number of microbial species present in the plant are inherent to the species and environmental conditions. The microbial biomass, species diversity and number of colony forming units at the time of ensiling determine the course of the fermentation process as well as the effectiveness of additives. In this context, the objective of this project is to characterize specific microbial population and its interaction with the presence of a microbial additive and relates these findings with silage fermentation profile in cane sugar silage. In the first experiment the microbial profile of sugar cane varieties IACSP93 and IAC91-2195-6006 untreated or treated with L. buchneri will be determined in the fresh forage, at the time of ensiling and after 90 days of storage. Total DNA will be extracted and amplified with primers V1-V3 region of the 16S ribosomal RNA (rRNA) in bacteria and ITS4 regions and ITS5 for fungi. Sequencing will be performed using a MiSeq (Illumina - San Diego, USA). The experiment will be completely randomized in a 2 × 2 factorial design, and means will be compared using the Tukey test (0.05). For the DNA libraries similarity test will be used and the products of the amplification reaction (amplicons) will be separated by electrophoresis on 1% agarose gel. In the second experiment L.buchneri will be quantified in varieties of cane sugar at the time of ensiling and at15, 30, 60, 75 and 90 days of storage using aq-PCR real time. The experiment will be completely randomized in a 2 × 2 × 5 factorial arrangement, and the averages will be analyzed using the Tukey test (0.05). (AU)
