Abstract

The manually extraction step, is a procedure that requires a large volume of samples, reagents and analysis time. The automation/mechanization aims to eliminate these limitations, and flow analysis systems are noted for serving these proposes with the use of minimum reagents volume, low cost, simplicity, reduced analysis time and minimization of the contamination risk. They are versatile systems, because they allow that samples or reagents are packed in columns, expanding the applications, e.g. extractions, solid phase reactions, separations, etc. However in systems with unidirectional and continuous flow, these have limitations such as backpressure effects, preferential pathways establishment and expansion or contraction effects. The use of pulsed flow and the establishment of fluidized bed condition can be an alternative to flow analysis systems proposed in the literature. The solenoid pump provide a turbulent mixing, improving the homogeneity of the chemical reaction and reducing the formation of preferential channels. In this context, the aim of this project is use pulsating pumps to fluidized beds establishment in a flow analysis system, to improve the efficiency of solid-liquid interaction, increasing the extraction and analytes adsorption processes in agronomic interest samples. The use of pulsed flows in studies involving soil and agronomic parameters seem to be pioneers. As specific objectives will be developed flow analysis systems for three experiments: 1) System for metals sequential extraction in soils; 2) System for determining the phosphorus adsorption capacity, remaining phosphorus (Prem) in soils; 3) System for determining cation exchange capacity (CEC) of soils, evaluating adsorption and extraction of the interest species. The procedures should provide significant improvements in analytical performance, especially the effluents volume generation and automation. (AU)
