Effect of energy restriction and genetic group on the microbiota of the gastrointestinal tract of broilers.

Abstract

This project aims to evaluate the effect of qualitative and quantitative dietary restriction and genetics on animal performance, carcass characteristics and major bacterial groups present in the digestive tract of broilers. Two experiments will be carried, the first with a commercial line of broilers, and the second with a slow growth line. All animals will be equality created until 21 days of age. At 21 days, they will be divided into 3 experimental treatments: control, restriction of 30% in ME (Metabolizable Energy) of 21 to 42 days and quantitative restriction of 70% of 21-28 days, followed by refeeding. The performance of the animals will be measured at 21 and 42 days. For microbiota analyses, will be collected 4 replicates from samples of the ileum and cecum at 21 and 42 days, and 28 days for treatments 1 and 3. For the analysis of absolute quantification by real-time quantitative PCR (qPCR), the metagenomic DNA of samples will be extracted and subjected to PCR amplification with specific primers for the bacterial groups to be studied, and compared with standard curves constructed from the plasmidial DNA of clones derived from the same fragments. The Ct values (Cycle Threshold) for each sample will be converted into the number of copies of 16S rRNA gene, and expressed as number of copies/g of mucus or intestinal contents.
