Abstract

Oral cancer is a highly incident disease in world population and it can cause facial deformities and death. The choice treatment is costly and usually involves the administration of chemotherapy, which may have cytotoxic action also on healthy cells, which affect significantly the quality and survival of patients. Thus, many researchers are working in discovering new molecules, which comes from natural sources that may have therapeutic interest. Curcumin, naturally occurring compound isolated from the rhizomes of Curcuma longa, it has shown activity in the treatment of oral cancer, it acts on the nuclear factor kappa beta (NF-kB), which mediates expression of genes that are involved in promoting tumors, angiogenesis and metastasis. However, its clinical use is facing many challenges, mainly due to their physicochemical characteristics and low oral bioavailability. Next, when incorporated in nanostructured systems precursors liquid crystal (SPLC) added of mucoadhesive polymer adjuvant, shows to be an interesting option to enable its use in the treatment of oral cancer, since these systems can protect the drug from degradation and to increase the residence time of the formulation in the oral environment, allowing a controlled release on an specific site of action. Furthermore, buccal administration of SPLC containing this drug can be advantageous, since the buccal mucosa has high permeability and high blood flow, allowing the local or systemic treatment and avoiding first pass metabolism, optimizing the local action. This project aims to develop a SPLC consisting of polyoxypropylene (5) polyoxyethylene (20) cetyl ether, oleic acid and polymer dispersion aqueous containing poloxamer associated with chitosan for buccal administration of curcumin and, subsequently, investigate the in vitro activity of formulations. Thus, we aim to develop a delivery system in nanostructural level for use in the treatment of oral cancer.
