Abstract

Serotonin (5-HT) is a neurotransmitter that regulates diverse physiological processes are involved in the adaptation to aversive events, such as forced immobilization. Serotonergic projections that depart from the Median Raphe Nucleus (NMnR) toward the Dorsal Hippocampus, would be responsible for adapting to stress. The 5-HT7 receptor is a 5-HT postsynaptic present in several brain structures, among them, the hippocampus and NMnR. Published data suggest that adaptation to stress involves the facilitation of serotonergic neurotransmission mediated by 5-HT7 in NMnR. The objective of this work is to study the role of 5-HT receptor type 5-HT7 in response to restraint stress. Therefore, the mice receive injections in intracerebral NMnR LP-44 and SB-258741 (5HT7 agonist and antagonist respectively) immediately before or after an event constraint forced acute lasting two hours and 24 hours after the animals will be submitted to test the elevated plus maze, which will be filmed for later behavioral analysis. Finally, there will be a transcardiac perfusion followed by histological analysis of the injection sites. Is used analysis of variance (ANOVA) and a two-way, according to the experiment. The results of this work will assist in understanding of the neurobiological mechanisms of major depressive disorder involving the participation or not of serotonergic neurotransmission mediated by 5-HT7.
