Abstract

Serotonin (5-HT) is a neurotransmitter that regulates various physiological and behavioral processes and is involved in adaptation to aversive events, such as forced detention and forced swimming. Serotonergic dysfunctions are associated to various disorders, such as depression. The serotonergic projections of the median raphe nucleus (MRN) towards the dorsal hippocampus, would be responsible for adapting to stress. The 5HT7 this is a 5-HT receptor in various brain structures, among them, the hippocampus and the MRN. Data from the literature and from our laboratory suggest that adaptation to stress involves the facilitation of serotonergic neurotransmission mediated by 5-HT7 in MRN. The objective of this study is to investigate the role of type 5-HT receptor 5-HT7 of MRN in response to restraint stress and the forced swimming. For this, Wistar rats with targeted cannula guide to MRN undergo a forced restriction acute event lasting two hours, receiving intra-MRN injections of LP-44 and / or SB-258741 (5HT7 agonist and antagonist, respectively) immediately prior (i) or after (ii) forced restraint according to the experimental condition. After 24 hours, the animals will be submitted to the test in the elevated plus maze, and their behavior filmed for later behavioral analysis. A tracking condition (iii) in which mice receive intra-NMnR treatment 24 hours before the test, without exposure to the stress constraint is performed. Similarly, mice that pass through the forced swim test are divided into three experimental conditions: i) receiving a pre-test immediately followed by intra-NMnR treatment LP-44 and / or SB-258741 and 24 hours later are submitted to the forced swimming test; ii) exposing the pre-test and 24 hours after intra-MRN then the test injection; iii) control group in which there will be exposure to pre-test and intra-NMnR injection followed by the test. Finally, the test animals are transcardially perfused submtidos followed by histological examination of the injection sites. Will be used analysis of variance (ANOVA) of one and two copies, according to the experiment. The results of this study will help us understand the neurobiological mechanisms of depression involving the participation or not of serotonergic neurotransmission mediated by 5-HT7 in MRN. (AU)
