Abstract

Major Depression is a mood disorders characterized by depressive mood, irritability, decreased self-steem, lack of hope, feelins of guilty, decreased concentration, eating and sleep disorders, and recurrent suicide thoughts. In the past few years, stress has been investigated as an important etiologic factor that leads to depression. It has been used as a tool in animal models of depression. In these studies, several compounds with antidepressant actions are being tested to see whether they attenuat or prevent the behavioral changes induced by stress.Among the brain structures involved in the neurobiology of depression, the hippocampus has received considerable attention. In this structure, facilitation of serotoninergic or blockage of NMDA-glutamatergice mediated neurotransmission are able to reverse or prevent the effects of stress. Dorsal Hippocampus receive most of its serotonergic afferent fibers from the Median Raphe Nucleus (MnRN) and the correct functioning of this pathway has been proposed as the main mechanism underlying behavioral adaptation to chronic stress. Lesions of MnRN serotoninergic neurons leads to significative behavioural changes in response to chronic stress. XXX, it is not clear whether the integrity of the MnRN-Dorsal Hippocampus pathway is essential to the antidepressant behavioral effects observed after treatment with different drugs such as imipramine in rats submitted to chronic stress. So, the aim of our work is to investigate if chronic treatment with the antidepressant imipramine can prevent the behavioral deficits induced by chronic exposure to restraint stress in animals with lesions in the MnRN-DH serotoninergic pathway.
