Evaluation of the effectiveness of different irrigation protocols in the root canal cleaning

Abstract

The microbial biofilms and their products are the main factors influencing the initiation, propagation and perpetuation of chronic apical periodontitis. The application of chemical solutions disinfectants, including chlorhexidine and sodium hypochlorite in different concentrations, are conventionally used in antisepsis of the root canal system. For this reason, irrigation protocols are employed using sodium hypochlorite or chlorhexidine associated with the use of acids and other substances. Thus the aim of this research is to evaluate, in Confocal Microscope Laser Scanning to evaluate the antimicrobial and scanning electron microscopy to evaluate the different protocols in removing biofilm andsmear layer. The protocols to be tested are: Sodium hypochlorite 1% and 2.5% EDTA associated to the peracetic acid or hydrogen peroxide 10volumes. Distilled water is used as a control. Each substance is in contact for 5 minutes. For the study of biofilm will be used blocks of bovine dentin sterilized, placed in a dental appliance that will be used by a volunteer for 3 days for oral biofilm formation. After biofilm formation, the samples will be evaluated in Confocal Microscope Scanning Laser for assessment of bacterial viability and scanning electron microscope in low vacuum to observe the formation of the biofilm where it will be analyzed in the block area of the biofilm.Subsequently the blocks will be submitted to the protocols according to the group. Are mpregados 5 blocks per group. After completion of the protocols, the blocks will be resubmitted in the analysis Microscope Laser Scanning Confocal and scanning electron microscopy and will analyze the remaining area of biofilm bacteria and the percentage of live and dead remnants. The difference of the area before and after antimicrobial and demonstrate the ability to eliminate biofilm of each protocol.To eliminate the smear layer, 50 teeth will be employed, which will be divided into 5 groups of 10 teeth each. The teeth are embedded in resinand screws are placed orientation. The teeth are cut longitudinally Isomet cutting machine. The teeth are mounted and instrumented to generate smear layer. After that the sections will be disassembled and analyzed by SEM. Later be reassembled and used different irrigation protocols. After the protocols, the teeth are removed and replaced analyzed by SEM. Consideration will be given to area smear layer before and after implementation of the protocol. The data will be compared statistically.
