Abstract

The Anticarsia gemmatalis multiple nucleopolyhedrovirus (AgMNPV) has been used as a biological pesticide for the control of soybean caterpillar, an important pest of soybean crops, in Brazil. Currently, AgMNPV is produced by infection of caterpillars (in vivo) and it has been studied the possibility of production by the infection of cells in culture (in vitro). In this direction, this project aims to study the loss of virulence and optimization of the parameters for the industrial production of the wild virus and its variant in in vitro systems.Studies will be made using the strain SF21 in static system cultures, and then in batch process in a bioreactor. After growth, the cells will be infected with the viral clone of baculovirus Anticarsia, AgMNPV 2D. Analyzes will be performed before and after the infection for growth of viable cells, substrate consumption (glucose and glutamate), lactate and polyhedra production and for the quality of the pesticide produced by morphology and DNA analysis.Analyzing the experimental data, it is expected to achieve greater knowledge about the molecular mechanisms that influences baculovirus in the host cells. Through the selection and isolation of viral clones Multiple Polyhedra (MP) more stable and from the kinetic parameters obtained from the experiments, it is believed that it will be possible to understand the loss of virulence, allowing, in this way, the production of biopesticide on a larger scale (bioreactor).
