Abstract

The study of the structure and genetic diversity in natural populations contains the description of the levels of genetic variation maintained within them, as well as the manner in which genetic variation is divided between them, thus identifying specific anthropogenic barriers which reduce genetic diversity and genetic flow. Thus, we can provide subsidies to help create significant units for conservation. Analyses of this scope can be performed with the use of molecular markers microsatellites, which are informative and currently represent the most polymorphic class known and used markers. This project aims to develop microsatellite markers for Trichocentrum pumilum (Orchidaceae) in order to be used in analyzes of spatial genetic structure, and small-scale genetic diversity and gene flow in natural populations of this epiphytic species that is widely distributed in North northeast, southeast, south and west-central Brazil. Three populations of T. pumilum will be sampled in Piracicaba , State of São Paulo , by collecting at least 15 individuals per population and georeferencing the position of each individual sampled. For the development of the methodology primers genomic library enriched with microsatellites will be used. With specific primers for T. pumilum, will be featured on the spatial genetic structure at small scale, and the genetic diversity of populations, and will be also evaluated the reproductive system and the intensity of gene flow species. This way, you can provide relevant information that can serve as input for management and conservation in the short, medium and long term programs. (AU)
