Evaluation of MMPs, TIMPs, RECK, SPARC and Bsg expression during rat ovarian dynamics

Abstract

The remodeling of the extracellular matrix (ECM) by MMPs, TIMPs, and RECK is directly related to embrionary development, angiogenesis, and organ morphogenesis. Specifically, in the ovaries, this remodeling allows the events sighted in the ovarian cycle, like the follicular growth, ovulation, and formation and regression of the corpus luteum. The resulting tissue organization is regulated, partially, by the action of proteolytic enzymes known as Matrix Metalloproteinases and their inhibitors, TIMPs. Currently, the remodeling of the ECM bu the MMPs/TIMPs system is strongly related to some genes' expression, such as Basigin (BSG), that induces the expression of MMPs during the ovarian cycle in rats, or the gene SPARC, that modulated angiogenesis and cell proliferation in several tissues, including ovary. Another gene involved in this system is RECK, already described in several cancers. The decrease in its expression together with an augmentation in the expression of MMPs has been associated with more aggressive and metastatic tumors. Recently, it was described the modulation of RECK by the hormone estrogen in the uterus, tissue in which tissue remodeling is controlled by reproductive hormones, as well as the ovaries. Even though RECK and SPARC are widely associated with tissue remodeling in several pathologies, their association with the naturally occurring MMPs/TIMPs-based ovarian tissue remodeling has not been described until now. Considering this, the hereby proposed work aims to analyze the genic and protein expression of some of the MMPs (2, 9, 12, 14, and 19), some of their inhibitors (TIMPs 1, 2, and 3, as well as RECK), SPARC and BSG during the natural and hormone-induced ovarian folliculogenesis throughout the estral cycle, with the objective to verify possible associations between those targets and ECM remodeling in the aforementioned processes.
