Abstract

The present study aims to investigate the microbial interactions and evaluate the role of surface adhesins of initial colonizers Streptococcus oralis and Actinomyces naeslundii in the formation and structure of Candida albicans biofilms. For this, Poly (methyl-methacrylate)resin discs will be fabricated and standardized according to the surface roughness, simulating the inner face of a prosthesis. The discs will be randomly distributed in a 24-well plate and given 1 ml of stimulated and filtered saliva for salivary film formation. Then, the discs will be transferred to a new 24-well plate in which occurs the formation of biofilms containing microorganisms Actinomyces naeslundii (An), Streptococcus oralis (So) and Candida albicans (Ca), divided into three groups: (I)biofilm containing An and Ca (II) containing So and Ca and(III)the biofilm containing the three species. Biofilms will be evaluated during adhesion (1.5 hours) and at 24 and 48 hours of development. Cell viability of each micro-organism will be determined by quantifying the polymerase chain reaction (qPCR) and by counting the colony forming units (CFU). The bacterial adhesins and protein co-aggregation CSH-like and Ag I/II will be investigated from assays that comprise the genetic modification of microorganisms, construction of mutants microorganisms and determining the presence of the adhesins by molecular assays. The structure and organization of biofilms will be evaluated with confocal laser microscopy and scanning electron microscopy (SEM).
