Abstract

The problem of water pollution acquires greater relevance in the context of a developing agrarian economy like Brazil. Even though, the leather industry is a important economic sector in Brazil, there is an increasing environmental concern regarding tanneries because they produce large amounts of potentially toxic wastewater containing dyes, which are equally hazardous for human population, aquaculture and agricultural activities in the area. This project proposes the development and validation of a qualitative method (confirmation) and quantitative based on liquid chromatography coupled to electrospray source - tandem mass spectrometry (LC-ESI-MS/MS) to be developed for determination of residues below level to assess contamination of tannery wastewater effluent in São Sebastião do Paraíso city, state of Minas Gerais. The target substance will Acid Black 210 dye. The extraction procedures will be optimized using solid phase extraction (SPE) with a cartridge Strata X. High performance liquid chromatography (HPLC) and electrospray ionization in negative mode (ESI) with multiple reaction monitoring (MRM) will have their parameters optimized and applied to real wastewater samples. The results will be analyzed and discussed to establish the relationship between the concentrations of the analyte and the toxicity of the dye Acid Black 210. It will be carry out in the application of the Comet Assay samples analyzed by LC-ESI-MS/MS in order to correlate the data to quantify the dye with biomarkers of xenobiotic effects of the action of the dye Acid Black 210 in water.
