Abstract

Obesity is now the major risk factor for a number of diseases, including metabolic syndrome and cardiovascular disease , including diabetes , atherosclerosis and hypertension . The activation of a subclinical inflammatory process in the hypothalamus has been shown to be the triggering factor of weight gain associated with disorders such as insulin resistance. The activation of this inflammatory process in obesity arises from a complex combination of genetic and environmental factors. The consumption of diets rich in saturated fatty acids is certainly one of the most important environmental factors , leading to inflammation through the activation of TLR4 receptors and endoplasmic reticulum stress. The involvement of the innate immune system in hypothalamic inflammation has been extensively studied. However, the role of adaptive immunity in the course of obesity is unknown. In neurodegenerative diseases, inflammation of the central nervous system is responsible for the attraction and activation of T lymphocytes, and this event concomitant to the onset of pathology. Studies have shown that intracerebroventricular administration of pro-inflammatory molecules such as TNF-±, promote the attraction and entry of lymphocytes in the hypothalamus. Given the importance of the immune system in the pathogenesis of obesity-related disorders , this study aims to evaluate the role of lymphocytes in the pathophysiology of hypothalamic obesity animal models. Through immunohistochemistry assays, Real Time - PCR and Western blot , we will analyze the contents of cells of the adaptive immune system in a time- course of obesity by feeding the animals with different time fat diet .
