Study of tylosin macrolide modulatory effect on acute inflammatory reaction in Nile tilapia (Oreochromis niloticus), vaccinated and challenged with Streptococcus agalactiae

Abstract

This study aims to evaluate the modulating effect of macrolide tylosin, administered by route oral in the diet, on the acute inflammatory response of tilapia , O. niloticus , vaccinated and challenged with S. agalactiae . For this study 133 Nile tilapia ( ± 300 g ) derived from the same spawning will be randomly placed in 19 aquaria ( n = 7 ) , with a capacity of 500L of water each , supplied with running water devoid of chlorine will be used from artesian wells, a flow rate of 1 L / min., providing the following treatments: V-T0= vaccinated and untreated with tylosin (n = 21) V- T5 = tylosin vaccinated and treated with 5mg/kg (n = 21 ), V = T10 - vaccinated and treated with tylosin 10mg / kg ( n = 21); NV- T0 = not vaccinated and untreated with tylosin (n = 21); NV- T5 = not vaccinated and treated with 5mg/kg tylosin (n = 21) and NV -T10 = not vaccinated tylosin and treated with 10mg/kg (n = 21); PS = Physiological standard (n = 7). After acclimatization period of 15 days, the bacterin of S. agalactiae containing 1.0 X109 cells per ml left dorsolateral be administered intramuscularly in the region at a dose of 0.5 mL . The vaccine stimulation will occur 15 days before the bacterial challenge in three replicates of groups V - T0 - T5 V and V - T10 . While the fish groups T0 - NV , NV - NV - T5 and T10 will be submitted to the administration of saline at the same dose, to standardize the stress of capture and handling . Feed containing tylosin will be taught for seven days prior to experimental challenge (treated groups with 5 and 10 mg of macrolide/ kg body weight) . After administration of bacterial inoculants in the swim bladder ( determined in a study of LD50) , the fish will be sampled post- challenge 6 , 24 and 48 hours ( HPD ) for blood , exudates and tissue for histopathological analysis . However , the correlation between studies of cell accumulation in inflammatory exudate, leukocyte count , leukocyte respiratory activity , determination of lysozyme activity and serum defensin , serum biochemistry and histopathology of hematopoietic organs will provide important information for assessing the modulatory capacity of the macrolide tylosin on acute inflammatory reaction of tilapia vaccinated and challenged with S. agalactiae .
