Abstract

The development of novel liposome formulations is crucial for enhanced drug delivery systems. In this Project, we shall prepare and characterize liposomes functionalized with polyethylene glycol (PEG) and the monoclonal antibody 2C5, which will be used as vehicles for the anti-cancer drug paclitaxel, widely used for treating breast and ovarian cancer. The major challenge is to incorporate paclitaxel in long-circulating immunoliposomes in order to increase the drug specificity and reduce its side effects. The latter include hypersensitivity reactions, nephrotoxicity and neurotoxicity. The drug delivery systems will be characterized by dynamic light scattering (DLS), zeta potential measurements, and transmission electron microscopy (TEM). To assess the cell interaction of the multifunctional liposomes, flow cytometry (FACS) analysis of HeLa and 4T1 cells incubated with liposomal formulations will be performed. It is hoped to understand the molecular-level interactions that may lead highly specific, effective drug delivery systems for cancer therapies. (AU)
