Abstract

The drugs used in the leukemia treatment, as Elspar® and Erwinase®, use L-asparaginase by prokaryotic origin and the main disadvantage of its use are hypersensitivity reactions, sometimes fatal. The use of L- asparaginase eukaryotic origin (yeast and filamentous fungi) can have lower intensity of allergic reactions due to post-translational properties that occur in these micro-organisms. Therefore, research on L-asparaginase stemmed micro-organisms eukaryotes proves to be relevant when taking into account the clinical condition of patients due to effects caused by the use of enzymes prokaryotic origin. Another issue that must be emphasized is that the lack of drug/biopharmaceuticals produced in Brazil, as the case of L-asparaginase, not only demonstrates problems in the area of public health, but also a problem of technological development in our country. We have examples of large-scale production of biotechnological products (ethanol, PLA) but the implementation of new industrial bioprocesses is still very modest in view of the increasing development of industrial microbiology. This is mainly due to poor integration of the various stages of the development of a bioprocess. The objective of this project is the optimization of culture conditions and the increase in the scale of production of extracellular L- asparaginase by Aspergillus terreus in bioreactors. This project emphasizes the need to develop steps in the development of bioprocesses, in order to rely on information to evaluate the technical and economic viability of the entire development process, and thereby assist in the consolidation of Enzyme Technology in Brazil and decrease the profile importer of this technology.
