Abstract

The aim of the present study will be evaluate the cytotoxicity of solvated and non-solvated adhesives with different amount of 2-hydroxyetil-metacrilate (HEMA), on dental pulp fibroblasts. The cells will be isolated from the dental pulp of sound human third molars. Model adhesive systems, solvated or not, will be manipulated with different concentration of HEMA (05, 10%, 20%, 30% and 40%). The fibroblasts 10 x 104 cells/well) will be seemed in sterile 6 wells dishes, and incubated. Cylindrical specimens (5mm diameter, 1 mm thick) will be prepared and maintained in contact with the culture medium for 6 h. After this, the obtained extracts (culture medium + products released from the adhesive system) will be applied on the cells and maintained in contact for 6 h. After this time, the extract will be removed and the cell metabolism will be analyzed using the metiltetrazolium (MTT assay). The obtained data will be tabulated and the statistical analysis will be performed.
