Abstract

The existence of a structure-function relationship in proteins is well known. Proteins evolved to fold into stable structures optimized for their functional roles. However, what can be the minimal information necessary in order to create a functional role? Different approaches have been proposed to address this issue. One of these approaches involves the concept of sparse spatial motifs to define the functional role of enzymes. In this approach, the atomic coordinates of a few amino acids are used to infer the enzyme activity. In this project we ought to test whether the insertion of sparse spatial motifs of a simple active site is sufficient to give functional elements to a different protein. For this purpose, we will look for protein structures that might serve as templates for the insertion of the spatial motif of a polysaccharide monooxygenase (previously known as glycoside hydrolase 61, GH61) containing two histidines and one tyrosine coordinating a cupper atom. The search for a template will be done using the software SMatch, developed in our group. The evaluation of the gain of activity will be done in two steps: the ability for binding the metal and the monooxygenase activity. (AU)
