Abstract

Fungal diseases represent a great challenge to the medical and dental areas, due to the increasing prevalence, few therapeutic options and antifungal resistance. Cold- plasma jet is a gaseous mixture containing charged particles, free radicals and radiation. Its potential application in infectious disease was reported, however there is still a lack of a systematic study on the antifungal effect and its mechanism of action. Moreover, studies on its genotoxic potential were not detected. The general aim of this project is to evaluate the antifungal effect of helium plasma jet on Candida albicans and Trichophyton rubrum and its genotoxic potential. The antifungal effect on planctonic cells and biofilms will be determined. Effects on fungal cells and on virulence factors will be evaluated, searching for evidences of the mechanisms of action. For C. albicans, assays for analysis of cell wall and cell membrane integrity, ergosterol quantification, morphogenesis, exoenzymes production, adherence to epithelial cells and in vivo effect in the treatment of oral candidosis in murine model will be performed. For T. rubrum, membrane and cell wall integrity, biomass production, micelial radial growth, cell morphology, keratinase activity and effects in an ex vivo nail infection model will be evaluated. The genotoxic potential will be evaluated in vitro on cultures of keratinocytes and CHO cells by using comet test. Data will be statistically analyzed, first by testing the normality of the sample distribution and then, by specific adequate analysis (±=5%). (AU)
