Abstract

The high incidence of candidoses caused by Candida albicans and its high adaptation and resistance capacity to antifungal boost the development of research to find alternatives therapies for this infection. Therefore, the goal of this paper is to evaluate the interaction between C. albicans and Lactobacillus rhamnosus, a probiotic bacterium, evaluating if the pathogenicity and the gene expression that set the biofilm formation of C. albicans, in vivo and in vitro. Will be used strains of C. albicans ATCC 18804 and L. rhamnosus ATCC 1465 from the Microbiology Laboratory of the São José dos Campos Institute of Science and Technology/UNESP. For this study will be made two suspensions of L. rhamnosus, one with a celular portion and other with only the culture supernatant, free of cells. Assays biofilm will be done in strains of 96 wells, where will be added both of the microorganisms for the interact between them. After 48 h the biofilms will be homogenized, diluted and seeded on sabouraud agar plates to count the colony-forming units (CFU/mL) of C. albicans. The metabolic evaluation of the C. albicans will be done by the colorimetric XTT assays, after the interaction in the biofilm model. The C. albicans genes will be quantified by real time RT-PCR after the interaction of this two microorganisms in the biofilm model using the proposed methodology. The experimental model Galleria mellonella will be used in the essays of the C. albicans pathogenicity in interaction with different L. rhamnosus suspensions. The results gain in this study will contribute for a better understanding between the interaction of this two microorganisms and it will be possible the application of probiotic bacteria in the development on new therapies in infection by Candida yeasts.
