Abstract The therapeutic semen cryopreservation, elective or for medical procedures such as radiotherapy, chemotherapy or invasive surgery, and also in assisted reproduction treatments, is of paramount importance. However, the conventional cryopreservation methods induce damage to sperm, associated with loss of motility and increase the sperm chromatin fragmentation. The conventional extenders medium based on egg yolk (test yolk buffer) contain additives of animal origin, which sets as less secure in terms of biosecurity, and its mechanism of action is not fully understood. In this respect, the new formulation of synthetic cryoprotective proposed in this project can, potentially, contribute to obtain accurate information about the action of cryoprotectants on the sperm cell and the development of safer strategies for semen cryopreservation. The objective of this project is to compare two media for cryopreservation of human semen, the conventional and synthetic supplemented with phospholipid and antioxidant, for the progressive sperm motility and sperm DNA fragmentation. In parallel, it will be evaluated the interaction of lipids present in cryoprotectant medium with the plasma membrane of the sperm. A clinical trial will be conducted, with randomized allocation of semen from the same patient for two parallel groups: control (ANTIOX-PC) and study (test yolk buffer). Quantitative parameters (semen analysis) and qualitative (TUNEL) of semen samples will be evaluated, before and after cryopreservation, in 63 volunteer patients recruited in the Infertility Clinic of the Division of Human Reproduction of Department of Gynecology and Obstetrics of HC-FMRP-USP.
