Abstract

Sirtuins are part of a class of enzymes that act in different cellular compartments with deacetylating histones and maintaining the condensed activating transcriptional regulatory DNA in the nucleus. Among the known activators of this enzyme , among the most efficient resveratrol , a polyphenol able to increase their rate of activity . Beneficial effects of resveratrol have been observed, and is of great importance to elucidate the molecular mechanism involving this substance, mainly in the liver, an important organ in glucose control by owning transcript factors closely related to several metabolic pathways and be the subject of common complications, as hepatic steatosis in uncontrolled diabetes. This study aims to investigate the effect of diabetic rats treated with resveratrol on related hepatic glycogen metabolism parameters. For this, will be investigated in diabetic rats (treatment with Alloxan) treated or not with resveratrol, hepatic glycogen content (ethanol extract), the mRNA expression of G6Pase, PEPCK, Slc2a2, and HNF- 1± (real time PCR) the abundance of SIRT1 and GLUT2 proteins and the percentage of binding of transcription factor HNF- 1± to the Slc2a2 gene. Expected to demonstrate that activation of SIRT1 improves hepatic glucose metabolism in diabetes.
