Effect of peracetic acid, used as irrigating solution, on the smear layer and on the penetration and bond strength of endodontic sealer to root dentin

Abstract

The aim of this study is to evaluate the effect of peracetic acid (PA), used as irrigating solution on the smear layer and the penetration and bond strength of an epoxy-based sealer to root dentin. For all tests, single-rooted extracted human teeth will be distributed into four groups (n = 10 per group) according to the irrigating solution used in biomechanical preparation: G1, AP 1%; G2, 2.5% sodium hypochlorite (NaOCl); G3, 2.5% NaOCl + 17% EDTA + 2.5% NaOCl and G4, saline. The canals of 40 roots will be instrumented with ProTaper rotary system until the F5. The specimens will be cleaved for evaluation of the smear layer on root dentin by a scanning electron microscope (SEM). To evaluate the effect of irrigating solutions on bond strength of endodontic sealer to dentin and its penetration in the root canal, the canals of 40 roots will be instrumented and irrigated in the same way that the SEM study. Root canals will be obturated with AH Plus manipulated with rhodamine B and the roots will be embedded in polyester resin, sectioned obtaining a specimen of each third of the root, which will be submitted to the push-out test. After the push-out test, the same specimens will be analyzed in inverted confocal microscope to assess the penetration of the sealer in root dentin. Data will be submitted to statistical analysis using the appropriate tests, with a significance level of 5%.
