In vitro evaluation of metabolic cell activity about different titanium alloys samples aiming for biomedical applications

Abstract

In recent decades there has been an increase in population life expectancy, resulting in growth in the use of metallic implants for biomedical applications. In addition to being biocompatible materials used to manufacture these implants must meet the appropriate modulus of elasticity and corrosion resistance. Titanium (Ti) and its alloys are the materials most commonly used for this purpose, since they exhibit such characteristics and elasticity can be controlled by making pores and the material from which the implant is made. The surface topography and surface energy of the material affect the biological mechanisms of the bone-implant interaction and their inflammatory responses. Thus, the main objective of this study is to evaluate the pattern of expression of pro-inflammatory cytokines, TNF -alpha and interleukin -6 , differentiation and osteoblast cell metabolism obtained from , opposite the samples made from different alloys of titanium, Ti6Al4V and Ti35Nb , showing porous surface . Samples will be made used by powder metallurgy , which will be divided into 2 groups: a) Group 1 : control - alloy Ti- 6Al -4V (titanium - aluminum -vanadium); b) Group 2: alloy Ti- 35Nb (niobium). Cells obtained from the cell line MG63, will be cultured for 3 and 10 days on the samples . Later on each sample, the tests will be performed to determine cell viability (MTT), quantification of the total protein, the protein carbonyl and the activity of alkaline phosphatase (ALP ), assessment of TNF- ± and IL -6 , and quantification of the nodule. All biological testing should be performed in three independent experiments. GraphPad Prism software (GraphPad, San Diego, CA) is used to perform statistical analyzes. The data will be analyzed using ANOVA. Where appropriate, comparisons will be made by the Tukey test. The significance level will be the standard value of 5 % .
