Chlorogenic acid effects during in vitro osteogenic differentiation of bone marrow-derived mesenchymal stormal cell of Wistar rats

Abstract

Osteoporosis is a metabolic bone disease essentially characterized by reduced bone mineral density associated with tissue microarchitecture damage. Epidemiologically relevant, this pathology has been showing increasing prevalence worldwide, which is in part explained by individuals increased life expectancy, worrying global public health organizations. Current treatments can be divided in anabolic therapies which aim to increase bone formation, and anti-resorptive therapies that reduce bone matrix resorption. Previous studies have shown a promoter effect of yerba mate (Ilex paraguariensis St. Hil. Var paraguariensis) on osteogenic differentiation, and chlorogenic acid has been reported as a major constituent of the extract, this information added to scientific literature data that already demonstrate clhorogenic acid negative effects on osteoclasts and their precursors, justify the evaluation of this compound in in vitro model of osteogenic differentiation, as seen the absence of studies about these effects. Therefore, the osteogenic differentiation will be induced in Wistar rats bone marrow-derived mesenchymal stromal cells with ascorbic acid, ²-glycerophosphate and dexamethasone associated with different concentrations of chlorogenic acid, for evaluation of following parameters, cell proliferation, total protein content, alkaline phosphatase activity and biological mineralization.
