Role of physical activity in benign prostatic hyperplasia and vesical dysfunction in rats with metabolic syndrome

Abstract

Introduction and objective: Sedentarism and obesity have been reported as relevant risk factors for the development of Benign Prostatic Hyperplasia (BPH) and vesical dysfunction. In the present work we investigated the role of physical activity in normal fed and high fat fed rats. Methods: Male Wistar rats with eight weeks old will be used in this experiment. This group will be randomly divided into four groups: 1. Physical Activity (PA) and normal diet; 2. Physical activity and high fat diet; 3. Sedentary and normal diet; 4. Sedentary and high fat diet. The rats in the groups 1 and 2 will submit to a swimming training protocol for 10 weeks, executed five times a week with duration of 60 minutes per day in a swimming system with warm water at 30-32°C. At the end of study, prostate glands and bladder will be dissected, weighted and stored for further analysis. We will measure the gene expression levels of insulin-like growth factor 1 (IGF1), AKT, MAPK, and genes related to tissue ischemia (VEGF, PDFG, HIF, and mTOR) through the quantitative real-time polymerase chain reaction (qRT-PCR) method, immunochemistry analysis of prostatic proliferation (ki-67, PCNA) and apoptosis (TUNEL), and bladder protein expression of TRIB3, AKT, Nos, GMPc, IGF1 and PKC trough western blotting analysis.
