Abstract

Contact dermatitis are considered complexes diseases and there is an estimative that they reaches about 15-20% of general population. Among cosmetic ingredients, UV-filters have been considered an emergent risk in skin allergy development and antioxidants, such as vitamin A can promote skin irritation. Most of tests used to assess skin sensitization still uses experimental animals, as LLNA - Local Lymph Node Assay, however there are many studies employing reconstructed skin model in international validation process. In Brazil, the availability of these techniques still presents some difficulties, as high cost and the need of an efficient system for importation of reconstructed skin models. Therefore, this study is relevant once it aims to assess the skin sensitization to UV-filters and vitamin A using a reconstructed skin model containing immune cells. For this purpose, two UV-filters and vitamin A will be applied in the reconstructed skin models containing immune cells and the amount of IL-1alpha, IL-17, IL-22 and IL-18 will be evaluated. The cell viability will be performed using MTT assay. Presence and differentiation of T-cells will be evaluated by transcriptomic analysis. Results will be compared to skin sensitization test performed in the EpiDerm"- 200 by measuring cell viability by MTT assay and quantifying IL-1±, and IL-18. Sodium dodecyl sulfate (SDS) and dinitrochlorobenzene (DNCB) will be used as irritant and sensitizer control. (AU)
