Influence of Hydrogen Peroxide on cell differentiation and mineralization of the pulp tissue after bleaching procedure. Histological and immunohistochemical study.

Abstract

Introduction: Previous studies of our research group indicate that hydrogen peroxide (H2O2) contained in the whitening gel can generate effects on the pulp tissue, which are not completely understood (Processo FAPESP nº 2013/25429-0, Cintra et al. 2013, Cintra et al. 2015, Cintra et al. 2015a). Among these effects are inflammation of mild to severe, protein denaturation, decreased cell metabolism, and necrosis (Camargo et al. 2007, Min et al. 2008, Costa et al. 2010, Cintra et al. 2013). Other studies show an induction to mineralization, leading to the subsequent calcification of large part of pulp tissue, and formation of nodules (Lee et al. 2006, Cintra et al. 2015). Objectives: To investigate in vivo the H2O2 penetration capacity in the pulp tissue by immunohistochemical staining of HO-1 and ROS; investigate the effects of whitening gel on dentinogenesis, by staining of Jun-D, OCN and OPN; and the influence of this gel on the activation of mesenchymal stem cells from pulp tissue by immunofluorescence technique, with concomitant identification positive for CD90, CD73, CD105 and negative for CD45. Methods: 60 rats will have the upper molars, the right or left side, bleached with 0,01mL H2O2 35% in a direct application of 30 minutes. Randomly, the molars side not cleared serve as a control. After 0 hours, 48 hours, 72 hours, 7 days, 15 days and 30 days (n = 10), animals will be killed and the jaws processed for histological, immunohistochemical and analysis imunofluorescência. Analysis: the tissue sections stained HE will be evaluated by predetermined criteria by assigning scores to inflammation, and results will be statistically analyzed by the Kruskal Wallis test and Dunn (p <0.05). After cell count, the values of ROS, HO-1, Jun-D, OCN, OPN, CD90, CD105, CD73 and CD45 obtained will be submitted to the Kolmogorov-Smirnov normality test and Shapiro-Wilk to define the statistical test to be used (p<0.05).
