Effects of incorporation of a bioglass on zirconia surface: evaluation of physical, chemical and biological proprieties

Abstract

Aiming the improvement of mucosa/implants interface, surface treatments have been proposed in order to direct and increase the cell proliferation around implants. The incorporation of bioactive materials on the abutments surface is an alternative to improve this interface. Therefore, the aim of the present study is to evaluate the effect of Biossilicate® incorporation on physical, chemical properties on the surface of zirconia discs and biological properties on the bioestimulation of oral mucosa cells. A Biossilicate® powder paste will be incorporated on zirconia sintered surface discs and they will be subjected to a heating process at 1100 ° C for 20 minutes to form a glassy layer on the surface. Then, the discs surface will be evaluated through SEM to observe the incorporation of bioglass and to check the distribution of the same on the specimens surface, and to identify of chemical elements with the aid of EDX. The alteration of color in samples also will be evaluated by spectrophotometer. The superficial roughness, contact angle, surface free energy will be determined. The materials will be sterilized and placed into wells of 24-well plates; the cells L929 and HaCat will be seeded onto the discs, separately on discs. No-incorporated zirconia discs and poliestirene discs will be used as controls groups, positive and negative, respectively. The cells proliferation (MTT assay) and morphology (SEM) will be evaluated at 1, 3, 5 and 7 days post-seeding. The migration capability of cells onto discs will be addressed by Wound Healing assay 24 hours after performing the "wound". Also, the collagen produced by L929 cells will be quantified after 7 days in culture onto the discs (Sirius Red). Data will be subjected to specific statistical analysis. (AU)
