Abstract

The organization of the oral bacterial species into biofilms is a challenge for the control of periodontal diseases, since bacterias aggregate in biofilms have a high resistance to antimicrobial agents. The use of systemic antibiotics in the treatment of these infections has demonstrated clinical benefits; however, the possible development of bacterial resistance justifies the search for other antimicrobial agents. Thus, the overall goal of this project is to test in models of complex biofilms in vitro the antimicrobial effects of a mouthwash containing 0.075% cetylpyridinium chloride, zinc and fluoride in an alcohol-free base. For that, we have as specific objectives to stablish this in vitro biofilm model with multiple periodontal species, mimicking the composition of dental plaque from subjects with periodontitis, and test in these biofilms the antimicrobial activity of different concentrations and exposition time to this mothwash. The device of Calgary Biofilm is used for the growth the biofilms. The composition of biofilms will be evaluated using the technique checkerboard DNA-DNA hybridization. Tests of inhibition of bacterial metabolism and time in biofilm inhibition of biofilm will be made for each solution tested. The statistical significance of differences between the various antimicrobial solutions will be tested using analysis of variance test for each exposure time separately. The significance of differences among groups will be tested using the Kruskall-Wallis and Dunn Test applying Bonferroni's correction for multiple comparisons. The significance of difference will be set at 95%.
