Abstract Bisphenol A (BPA) is a chemical compound found in products based on epoxy resins and polycarbonate plastics. BPA has been investigated by its activity on changing breast development and susceptibility to carcinogenesis when given on prenatal and/or postnatal stages. Gestational protein restriction (GPR) is related to changes on breast development of the offspring and the susceptibility to breast cancer in adulthood. Thus, this research project aims to: (1) assess whether BPA is able to intensify the adverse effects of GPR on the development of the mammary gland on female offspring ; 2) evaluate the effects of the GPR association and prepubertal exposure to BPA on morphological, immunohistochemical and molecular parameters in the mammary gland of female offspring on postnatal day (PND 51); 3) evaluate the effects of the association to GPR and prepubertal exposure to BPA on the susceptibility to breast cancer induced by administration of 7,12-dimethylbenz (a) anthracene (DMBA) in the offspring of females in the PND 180. Thus, pregnant females Sprague-Dawley strain will be divided into four groups: G1: normal protein diet (NPD); G2: low protein diet (LPD); G3: NPD + BPA (5x 250¼g / kg per week PND 21-51, administered by gavage) and G4: LPD + BPA. At PND 51, some of females will be euthanized for evaluation of the adverse effects of BPA in combination with GPR. The other females will receive a single dose of DMBA (30mg/kg by gavage) for the development of mammary tumors. Females initiated with DMBA will be sacrificed on PND 180. The glands and mammary tumors will be analyzed for incidence, morphology, histological features, immunohistochemistry and molecular analysis. (AU) | |
