Abstract

Orange rust of sugarcane is caused by Puccinia kuehnii, a recent foliar pathogen in Brazilian sugarcane fields, but already among the major diseases of the crop. Understanding the behavior of such disease is very important in comprehending its mechanisms of infection. This project aims to assess the transcriptome of the plant-pathogen interaction between sugarcane and P. kuehnii, during the establishment and development of the disease, by RNA sequencing using a next generation sequencing platform. The biological material to be used corresponds to sugarcane leaves from the susceptible cultivar SP89-1115 infected with P. kuehnii, sampled at 0h, 12h, 24h, 48h, 5 days and 12 days after infection; each time point was represented by three biological replicates, except for the initial time (control), for which we obtained two replicates. The expression levels of each transcript, in each of the sampling times, will be employed to detect differentially expressed genes with the R package DESeq2. These results will also allow the clustering of transcripts with similar expression patterns, i.e., increasing, decreasing or constant expression level throughout the infection period. Finally, we will perform an evaluation of the relevant metabolic pathways with BLAST2GO, in order to verify the occurrence of their modulation as the pathogen advances on the sugarcane leaves. (AU)
