In vivo and in vitro effect of extravirgin oil in the metabolism of osteoblastic cells from bone marrow of ovariectomized rats

Abstract

Osteoporosis is a disease with high incidence in older ages as well as it affects the most women in the postmenopausal period. This disease is associated to hormonal adjustments, genetic susceptibility and poor habits such as sedentary lifestyle and improper diet. It has been studied in order to find a viable form of treatment and particularly its prevention. In such way, alternative therapies to osteoporosis prevention and treatment are increasing in the last few years, mainly with the aid of functional foods with antioxidative properties like extravirgin olive oil (EOO). Thus, the goal of this study is to perform in vivo and in vitro studies to evaluate the metabolism of osteoblastic cells from bone marrow of ovariectomized rats in the presence of EOO. There will be used wistar female rats, which will be submitted to ovariectomy; immediately after the surgery, the selected groups will receive 1ml/100g body weight by gavage of EOO for 60 days and then sacrificed for removal of both femurs for cell isolation, which will be grown in flasks in osteogenic medium until subconfluence and seeded at a concentration of 2 x 104 cells per well in culture plates (n = 5). After the experimental period, the following parameters will be evaluated: quantitative and qualitative histological evaluation (collagen volume), cell proliferation and viability, alkaline phosphatase activity and detection, total protein content, quantification of mineralized nodules. The results will be analyzed by statistical programs consistent with significance p d 0,05, with the goal of finding statistically significant data showing the beneficial effects of extravirgin olive oil.
