In vitro effect of yerba mate in the metabolism of osteoblastic cells from bone marrow of ovariectomized rats

Abstract

Osteoporosis after menopause is characterized by a decrease in mineral bone density, resulting in reduced force and injuries in trabecular bone. Several therapies have been evaluated, mainly the utilization of Drugs, which may lead to side effects. In such way, alternative therapies to osteoporosis prevention and treatment are increasing in the last few years, mainly with the aid of functional compounds with antioxidative properties like yerba mate (ilex paraguariensis). Thus, the goal of this study is to perform in vivo and in vitro studies to evaluate the metabolism of osteoblastic cells from bone marrow of ovariectomized rats that received mate tea. There will be used wistar female rats, which will be submitted to ovariectomy; immediately after the surgery, the selected groups will receive 20 mg/kg body weight by gavage of yerba mate diluted in water until sacrifice. After 60 days, both femurs will be collected for histological analysis and cell isolation, which will be grown in flasks in osteogenic medium until subconfluence and seeded at a concentration of 2 x 104 cells per well in culture plates (n = 5). After the experimental periods, the following parameters will be evaluated: quantitative and qualitative histological evaluation, cell proliferation and viability, alkaline phosphatase activity and detection, total protein content, as well as quantification of mineralized nodules. The results will be analyzed by statistical programs consistent with significance p d 0,05, with the goal of finding statistically significant data showing the possible beneficial effects of yerba mate.
