In vivo and in vitro effect of grape seed extract in bone metabolism and in osteoblastic cells from bone marrow of ovariectomized rats

Abstract

Osteoporosis is a major concern in public health, mainly affecting elder people and women after menopause. Hormonal replacement shows positive effects on bone metabolism but may promote side effects such as breast and uterine cancer. An alternative would be the use of natural substances like grape seed extract (GSE), which has a great antioxidant potential for containing proanthocianidines (PACs). Thus, the purpose of this investigation is to evaluate by means of in vitro and in vivo experiments if GSE is capable of stimulate functional activity and impair bone loss in rats submitted to osteoporosis. There will be utilized 42 rats Wistar Hannover divided in 3 groups: sham, ovariectomized (OVX) and ovariectomizado/GSE (OVX/GSE). A concentration of 250 mg/Kg body weight of GSE (or distilled water for sham and OVX groups) will be administered by gavage for 60 days immediately after ovariectomy. After this period, the animals will be euthanized and femurs collected to perform qualitative and quantitative histological analysis. Besides, bone marrow mesenchymal cells will be isolated to be differentiated in osteoblasts to analyze cell viability, ALP activity and in situ detection, as well as mineralized nodules. Data obtained will be analyzed by adequate statistical test for p d 0.05, with the goal of finding the positive effects of GSE in bone metabolism.
