Effects of glucosamine sulfate and chondroitin sulfate compound and LLLT associated or not the articular cartilage in transection model of anterior cruciate ligament in rats

Abstract

Osteoarthritis (OA) is characterized as a chronic progressive disease worldwide frequent being the most affected knee joint. The compound of glucosamine sulphate and chondroitin sulphate and the low level laser therapy (LLLT- Low-Level Laser Therapy) has been frequently used as a non-invasive therapeutic strategies to alleviate and prevent the development of OA, however the signaling pathways driven by these resources remain uncertain. Thereby, the objective of this study is to measure the effects of the compound of glucosamine sulfate and chondroitin sulfate, the LLLT and the combination of both on the articular cartilage of rats knee subjected to induced model of OA through the transection of the anterior cruciate ligament (ACLT). Will be used 40 Wistar rats, males, divided into 4 groups (n = 10): osteoarthritic (OA); osteoarthritic and submitted to LLLT (OAL); osteoarthritic and subjected to treatment with the compound of glucosamine sulphate and chondroitin sulphate (OAGC); osteoarthritic and subjected to treatment with the compound of glucosamine sulphate and chondroitin sulphate associated with LLLT (OALGC). The treatments will begin 24 hours after surgery for 30 consecutive days. A combination of glucosamine sulfate 500 mg / kg of chondroitin sulfate and 400 mg / kg (orally). Moreover, a laser is used AsGaAl (continuous; » = 808 nm, P = 50 mW; spot area = 0.028 cm2; t = 28 s; D = 50 J / cm2; E = 1.4 J; irradiance = 17, 8 mW / cm2) in 2 points of the left hind leg (medial and lateral joint line) will be administered. To evaluate and compare the effects of treatment, histological analyzes will be performed using the criteria of OARSI, morphometric thickness and density of chondrocytes, as well as analysis of protein expression of IL-1² and MMP-13.
