Subtractive immunization efficacy on production of antibodies specific to prostate tumor cells

Abstract

Prostate cancer (PCa) is the second most prevalent neoplasm amongst men. Treatments grant survival, however they do not always cure; making it appropriate the adoption of adjuvant therapies, such as the passive immunotherapy with the use of tumor-specific monoclonal antibodies (mAbs). The mAbs originated from conventional immunization recognizes immunodominants epitopes. The subtractive immunization (SI) aims to raise the production of mAbs against poorly immunodominant antigens, capable of distinguishing between similar targets. This current project aims to assess the efficacy of the SI to obtain murine mAbs able to recognize tumor cells exclusively. For the immunizations, two human prostate cell lines will be used: RWPE-1 (non-tumoral) and PC3 (tumoral). Two groups of mice BALB/C (n=5) will be immunized; group 1 will receive the conventional immunization (PC3 solely) and group 2 will be subject to SI (RWPE-1, followed by cyclophosphamide and PC3). After the cell fusion, the screening of the hybridomas will be performed. The antibodies obtained will be tested against both cell lines. To analyze the effectiveness of the proposed technique in comparison to its conventional counterpart, it will be assessed: 1)Number of hybridomas obtained; 2)Number of positive hybridomas; 3) Number of hybridomas secreting antibodies of reactivity only to tumor cells and 4) Number of hybridomas secreting antibodies with potential use in immunocytochemistry.
