Abstract

Parkinson's disease (PD) is recognized as the second most common neurodegenerative disease in the population. Currently, the DP has no cure; however, the symptoms can be minimized by treatment with new drugs such as cannabidiol. Anandamide and 2- araquidonoglicerol, are endogenous cannabinoids derived from arachidonic acid and they bind to the CB1 and CB2 receptors, acting as agonists. Recently, animal studies suggest a possible change level of these substances in metabolism of animal used as models of PD. However, such studies involving quantification of these compounds in order to elucidate the biochemical this disease are still at an early stage. In recent years, liquid chromatography coupled to tandem mass spectrometry electrospray ionization (ESI) (LC-MS / MS) has become the reference analytical technique for analyzing various substances in biological fluids. The sample preparation step has been required for the development of chromatographic methods to remove interfering compounds and pre-concentrate analytes, often present at trace levels in biological samples. Current analytical chemistry has been directed to simplification by hyphenation techniques, automation (high-throughput performance) and miniaturization of the analytical systems. In this context, we highlight the column switching liquid chromatography coupled to tandem mass spectrometry (LC-MS / MS in column switching mode). The development of stationary phases for the first dimension (1D) of the analytical system, such as polymerizable ionic liquid (PIL) allow direct introduction of biological fluids in chromatographic systems and conduct a selective pre-concentration of the analytes. The great potential of the PIL as sorbent is attributed to its excellent chemical and physicalstability, great sorption capacity and selectivity, which can be adjusted by changes in the structure of the monomer synthesis. In this study, PIL will be synthesized, characterized by different spectroscopic techniques and chemically immobilized on the inner walls of fused silica capillaries. These capillaries will are evaluated as columns 1D LC-MS / MS system in column switching method for the determination of endocannabinoids in plasma samples of patients with PD disease undergoing therapy cannabidiol.
