Abstract

The burn is defined as a traumatic injury of thermal origin which affects the organic tissue and can be classified according to the skin layer reached. The higher the initial injury, the more severe the depth and extent. Taking into account the complexity of the tissue repair process, the treatment of burns always been a challenge, both for its severity, as the multiplicity of complications that commonly occur. According to the literature, using LLLT to treat burns, especially in the early stages of the process, it promotes better healing and less tissue repair time. Another area that is being investigated for the treatment of burns and wounds is the use of the application of stem cells, especially mesenchymal cells. Thus, it is believed that the combination therapy would be both capable of accelerating the burn healing process, thereby reducing the time and possible complications internment. The objective will be to investigate the effect of low level laser therapy (660nm)(LLLT) with or without treatment with mesenchymal stem cells (MSCs) in third degree burn. For that will be realized two experiments, one "in vitro" that will target the action of different fluencies of LLLT (10 J/cm², 20 J/cm² and 30 J/cm²) on the proliferation of MSCs and a study "in vivo" the what better fluency in the previous experiment will be used for the application in animals subjected to third degree burn. The "in vivo" study will be conducted in 96 rats Wistar, divided into four groups: GC: control group, GL: animals treated with LLLT (660nm), GCT: animals treated with MSCs and GLCT: animals treated with the combination of LLLT (660nm) and MSCs. The treatment of groups that use LLLT will be held three times a week, every other day for 30 days and the total experiment "in vivo". Groups that will be treated with MSCs receive ten applications of cells in total volume of 2000¼L. It will be done after the photographic monitoring induction of the burn and at 7, 15, 21 and 30 days after the start of the experiment to assess the degree of contraction of the lesion. The animals will be sacrificed at four distinct periods (7, 15, 21 and 30 days after the experimental procedure) being held collection of tissues for further histological and immunohistochemistry analysis. Statistical analysis will be performed to test the analysis of variance, two-way ANOVA. The level of significance is 5%.
