Abstract

Hematopoiesis is the process in which cells from the lymphatic and hematopoietic systems are formed, starting from precursor cells that have multipotency and self-renewal capabilities - the hematopoietic stem cells (HSCs). Appropriate mature progeny maintenance and production requires numerous cellular mechanisms related to HSC proliferation and differentiation, where cytokines act as direct modulators. Alterations in such processes may lead to cellular dysfunctions and the development of a leukemic phenotype, characterizing the leukemic stem cells (LSC). Recently, our group showed the ubiquitous Ca2+ signaling role in HSC and LSC proliferation, differentiation as well as cell death, yet, how small and oscillatory variations in intracellular Ca2+ concentrations, elicited by cytokines, modulate HSC responses is still unknown. Wnt signaling has been recently associated with HSC fate regulation and intracellular Ca2+ variations. Therefore, we aim to investigate the crosstalk between Ca2+ and Wnt signaling during HSC and LSC fate, using techniques, such as single cell analysis and transfection of HSC and LSC.
