Effect of gutta-percha on in vitro biomodulation of periodontal ligament fibroblasts

Abstract

Gutta-percha is stablished as root canal filling material due to its physical properties and biocompatibility. The addition of bioglass has being proposed in order to improve gutta-percha properties, however the biomodulation effect of bioglass containing-gutta-percha is still unknown. Taking this into consideration, this project aims to investigate the biomodulation of bioblass containing gutta-percha cones on periodontal ligament fibroblasts in vitro. Extracts of conventional and bioglass containing gutta-percha cones will be prepared in culture medium and serially diluted in order to be tested for cytotoxicity on human periodontal ligament fibroblasts cultures after 24, 48 and 72 h by using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT). The lower dilution that did not compromise cell viability will be employed for new stimulation for 72 h for RNA extraction. The samples will be subjected to RT-qPCR for detection of the inflammatory mediators TNF-±, IL-6 and COX-2 besides collagen 1 and cement protein 1. Calcium and phosphorus content will be determined by means of colorimetric assays. Data will be submitted to statistical analysis by means of ANOVA followed by Tukey test.
