Abstract

The role of thyroid hormone on testicular development is well described in mammals, however in fish is poorly explored. Considering that in mammals most of the experimental studies to date are focused on thyroid hormone effects on the developing testes, only limited data are available on adult testis and spermatogenesis. Therefore, the aim of this study is to evaluate the effects of thyroid hormone on zebrafish testis function and spermatogenesis. First, males will be exposed with 1 mM methimazole, a thyroid hormone disruptor for 21 days. Next, testes will be dissected out and cultured using an ex vivo organ culture system to study the effects of T3 on androgen release, spermatogenesis, gene expression and caspase activity. Spermatogenesis will be quantified evaluating cyst frequency and germ cell proliferation. The results of this study will provide contributions to teleost reproductive physiology and to comparative endocrinology. We expect to clarify the mechanisms how thyroid hormone act in the spermatogenesis and testis function. (AU)
