Abstract

Several baculoviruses have been used as biological pesticides for the control of soybean caterpillar, an important pest of soybean plantations in Brazil, such as Anticarsia gemmatalis multiplenucleopolyhedrovirus (AgMNPV). Currently, AgMNPV is produced through caterpillar infection (in vivo) and the possibility of production by insect cell infection in culture (in vitro) is studied. To do so, it is necessary to determine the best cell culture conditions that will be employed in the production of the virus, in order to maximize the production of the viral polyhedra. In this direction, the present project has the objective of studying the process of baculovirus production in Sf-9 insect cells for the production of bioinsecticide. For this, important factors such as the growth of insect cells in bioreactors will be taken into account to obtain cultures with high cell density. The nutrient supply to the cells will be tested in different ways, through fed-batch cultures, in order to prolong the exponential growth phase, resulting in a higher cell concentration to be infected. The infection stage will also be studied, obtaining parameters to guarantee the virulence of the product (baculovirus polyhedra). Analyzes of viable cell growth, substrate consumption (glucose and glutamate) and lactate production will be performed to monitor cell development. Analyzing the experimental data, it is expected to better understand cell growth metabolism and thereby improve the infection process and cultures in bioreactors. (AU)
